The Val637Ile variant has not been reported in the literature nor previously ide ntified in 369 individuals tested by our laboratory. Valine (Val) at position 6 37 is not 100% conserved across different species (opossum has a glutamine, chic ken has a leucine, tetraodon has a methionine), reducing the likelihood that the change is pathogenic. In addition, the ABCC9 gene has not yet been linked to l eft ventricular noncompaction. In summary, the currently available evidence sugg ests that the Val637Ile variant is unlikely to be pathogenic but additional stud ies (healthy control and/or segregation studies) are required to determine its s ignificance with certainty. -

This sequence change replaces valine, which is neutral and non-polar, with isoleucine, which is neutral and non-polar, at codon 637 of the ABCC9 protein (p.Val637Ile). This variant is present in population databases (rs113542001, gnomAD 0.05%). This variant has not been reported in the literature in individuals affected with ABCC9-related conditions. ClinVar contains an entry for this variant (Variation ID: 45393). An algorithm developed to predict the effect of missense changes on protein structure and function outputs the following: PolyPhen-2: "Benign". The isoleucine amino acid residue is found in multiple mammalian species, which suggests that this missense change does not adversely affect protein function. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -
