The encoded protein can promote tumor formation by targeting tumor suppressor proteins, such as p53, for proteasomal degradation. This gene is itself transcriptionally-regulated by p53. Overexpression or amplification of this locus is detected in a variety of different cancers. There is a pseudogene for this gene on chromosome 2. Alternative splicing results in a multitude of transcript variants, many of which may be expressed only in tumor cells. [provided by RefSeq, Jun 2013]

Experimental studies are conflicting or provide insufficient evidence to determine the effect of this variant on MDM2 protein function (PMID: 23653682). This variant has not been reported in the literature in individuals with MDM2-related conditions. This variant is not present in population databases (ExAC no frequency). This sequence change replaces threonine with alanine at codon 16 of the MDM2 protein (p.Thr16Ala). The threonine residue is highly conserved and there is a small physicochemical difference between threonine and alanine. -
