This sequence change replaces glycine, which is neutral and non-polar, with serine, which is neutral and polar, at codon 59 of the GJB2 protein (p.Gly59Ser). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individuals with Vohwinkel syndrome and Bart–Pumphrey syndrome (PMID: 15952212, 17106596, 30565282). ClinVar contains an entry for this variant (Variation ID: 17034). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt GJB2 protein function.
