The c.1498G>A (p.G500S) alteration is located in exon 10 (coding exon 9) of the BRCA2 gene. This alteration results from a G to A substitution at nucleotide position 1498, causing the glycine (G) at amino acid position 500 to be replaced by a serine (S). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. -

In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt BRCA2 protein function. This variant has not been reported in the literature in individuals affected with BRCA2-related conditions. This variant is not present in population databases (ExAC no frequency). This sequence change replaces glycine with serine at codon 500 of the BRCA2 protein (p.Gly500Ser). The glycine residue is weakly conserved and there is a small physicochemical difference between glycine and serine. -
