Three of five in-silico tools predict a benign effect of the variant on protein function. The variant allele was found at a frequency of 2.8e-05 in 251204 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.2274T>G has been reported in the literature in individuals affected with breast cancer (examples: Fernandes_2016, Guindalini_2022 ) or upper tract urothelial carcinoma (Vlachostergios_2020). These reports do not provide unequivocal conclusions about association of the variant with Hereditary Breast And Ovarian Cancer Syndrome. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publications have been ascertained in the context of this evaluation (PMID: 27741520, 35264596). The variant was also identified in dbSNP (ID: rs142243359) as "With Likely benign, Uncertain significance allele", ClinVar (classified as benign by Ambry Genetics; as uncertain significance by Invitae, GeneDx and two clinical laboratories), and in LOVD 3.0 (2x). The variant was not identified in UMD-LSDB database. The variant was identified in control databases in 10 of 277004 chromosomes at a frequency of 0.00004 (Genome Aggregation Database Feb 27, 2017). The variant was observed in the following populations: African in 8 of 24030 chromosomes (freq: 0.0003), Latino in 2 of 34376 chromosomes (freq: 0.00006), while the variant was not observed in the Other, European, Ashkenazi Jewish, East Asian, Finnish, or South Asian populations.
