Genes affected
BRCA2 (HGNC:1101): (BRCA2 DNA repair associated) Inherited mutations in BRCA1 and this gene, BRCA2, confer increased lifetime risk of developing breast or ovarian cancer. Both BRCA1 and BRCA2 are involved in maintenance of genome stability, specifically the homologous recombination pathway for double-strand DNA repair. The largest exon in both genes is exon 11, which harbors the most important and frequent mutations in breast cancer patients. The BRCA2 gene was found on chromosome 13q12.3 in human. The BRCA2 protein contains several copies of a 70 aa motif called the BRC motif, and these motifs mediate binding to the RAD51 recombinase which functions in DNA repair. BRCA2 is considered a tumor suppressor gene, as tumors with BRCA2 mutations generally exhibit loss of heterozygosity (LOH) of the wild-type allele. [provided by RefSeq, May 2020]

Pathogenic, criteria provided, single submitterclinical testingGeneDxFeb 12, 2018This deletion of two nucleotides is denoted BRCA2 c.2636_2637delCT at the cDNA level and p.Ser879Ter (S879X) at the protein level. The normal sequence, with the bases that are deleted in brackets, is GACT[delCT]GAAG. The deletion creates a nonsense variant, which changes a Serine to a premature stop codon. This variant is predicted to cause loss of normal protein function through either protein truncation or nonsense-mediated mRNA decay. Using alternate nomenclature this variant has been defined as BRCA2 c.2634delCT, BRCA2 2864_2865delCT, and BRCA2 2864delCT. BRCA2 c.2636_2637delCT has been observed in association with hereditary breast and ovarian cancer (Thirthagiri 2008, Toh 2008, Heramb 2018). This variant is considered pathogenic. -
Pathogenic, criteria provided, single submitterclinical testingQuest Diagnostics Nichols Institute San Juan CapistranoApr 17, 2024The BRCA2 c.2636_2637del (p.Ser879*) variant alters the translational reading frame of the BRCA2 mRNA and causes the premature termination of BRCA2 protein synthesis. This variant has been reported in the published literature in individuals/families affected with breast and/or ovarian cancer (PMIDs: 18431501 (2008), 29339979 (2018), 29884136 (2018), 35464868 (2022)). It was also identified with a second variant (BRCA1 c.4186-?_4357+?dup) in one individual affected with breast cancer and one reportedly healthy individual, however, the information available for this co-occurrence is limited (PMID: 27836010 (2016)). This variant has not been reported in large, multi-ethnic general populations (Genome Aggregation Database, http://gnomad.broadinstitute.org). Based on the available information, this variant is classified as pathogenic. -
Hereditary breast ovarian cancer syndrome Pathogenic:2
Pathogenic, criteria provided, single submitterclinical testingLabcorp Genetics (formerly Invitae), LabcorpJan 21, 2023For these reasons, this variant has been classified as Pathogenic. ClinVar contains an entry for this variant (Variation ID: 51318). This variant is also known as c.2634delCT. This premature translational stop signal has been observed in individual(s) with breast and/or ovarian cancer (PMID: 18431501, 26187060, 27836010, 29339979). This variant is not present in population databases (gnomAD no frequency). This sequence change creates a premature translational stop signal (p.Ser879*) in the BRCA2 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in BRCA2 are known to be pathogenic (PMID: 20104584). -

Hereditary cancer-predisposing syndrome Pathogenic:1
Pathogenic, criteria provided, single submitterclinical testingAmbry GeneticsAug 25, 2021The c.2636_2637delCT pathogenic mutation, located in coding exon 10 of the BRCA2 gene, results from a deletion of two nucleotides at nucleotide positions 2636 to 2637, causing a translational frameshift with a predicted alternate stop codon (p.S879*). This alteration has been identified in individuals diagnosed with breast and/or ovarian cancer (Toh GT et al. PLoS One, 2008 Apr;3:e2024; Pajares B et al. BMC Cancer, 2018 Jun;18:647). This alteration was also identified in two individuals who also carried a BRCA1 mutation (c.4186-?_4357+?dup) (Rebbeck TR et al. Breast Cancer Res, 2016 11;18:112) and in a large, worldwide study of BRCA1/2 mutation positive families (Rebbeck TR et al. Hum Mutat, 2018 05;39:593-620). This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD). Of note, this alteration is also designated as c.2634delCT in published literature. In addition to the clinical data presented in the literature, this alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation. -
