Truncations downstream of this position have been classified as pathogenic by our laboratory (eg. c.3226_3230delGTAGT, p.Val1076fsX3; c.3264dupT, p.Gln1089fsX10; c.3362C>G, p.Ser1121X). The variant was absent in 119496 control chromosomes (ExAC). The variant, c.3217C>T, has been reported in the literature in individuals affected with Hereditary Breast and Ovarian Cancer (Lang_2017, Sun_2017). These data indicate that the variant is likely to be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. Three clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014 without evidence for independent evaluation and classified the variant as pathogenic. This changes the amino acid from a glutamine to a stop codon within coding exon 10. This mutation was identified in 1/2991 Chinese breast cancer patients and in 0/1043 healthy controls (Lang GT et al. Int. J. Cancer 2017 Jul;141:129-142). In addition to the clinical data presented in the literature, this alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation. - |
