Hereditary cancer-predisposing syndrome Uncertain:2Benign:1
Missense variant in a coldspot region where missense variants are very unlikely to be pathogenic (PMID:31911673). -
The p.T1505A variant (also known as c.4513A>G), located in coding exon 10 of the BRCA2 gene, results from an A to G substitution at nucleotide position 4513. The threonine at codon 1505 is replaced by alanine, an amino acid with similar properties. This alteration was detected in a cohort of 210 male breast cancer patients and classified as likely neutral by the authors (Ding YC et al. Breast Cancer Res Treat. 2011 Apr;126(3):771-8). This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
This variant is located in the BRCA2 protein. Computational prediction suggests that this variant may not impact protein structure and function (internally defined REVEL score threshold <= 0.5, PMID: 27666373). To our knowledge, functional studies have not been reported for this variant. This variant has been reported in individuals affected with breast cancer, including one male individual (PMID: 20927582, 33471991; Leiden Open Variation Database DB-ID BRCA2_007370). This variant has been identified in 1/250254 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance. -

This variant is denoted BRCA2 c.4513A>G at the cDNA level, p.Thr1505Ala (T1505A) at the protein level, and results in the change of a Threonine to an Alanine (ACC>GCC). Using alternate nomenclature, this variant would be defined as BRCA2 4741A>G. This variant was observed in at least one individual with male breast cancer (Ding 2011). BRCA2 Thr1505Ala was not observed in approximately 6,500 individuals of European and African American ancestry in the NHLBI Exome Sequencing Project, suggesting it is not a common benign variant in these populations. Since Threonine and Alanine differ in polarity, charge, size or other properties, this is considered a non-conservative amino acid substitution. BRCA2 Thr1505Ala occurs at a position that is not conserved and is located in the RAD51 binding domain and POLH binding domain (Roy 2012, Buisson 2014). In silico analyses are inconsistent regarding the effect this variant may have on protein structure and function. Based on currently available evidence, it is unclear whether BRCA2 Thr1505Ala is a pathogenic or benign variant. We consider it to be a variant of uncertain significance. -
Hereditary breast ovarian cancer syndrome Uncertain:1
This sequence change replaces threonine, which is neutral and polar, with alanine, which is neutral and non-polar, at codon 1505 of the BRCA2 protein (p.Thr1505Ala). This variant is present in population databases (rs80358682, gnomAD 0.0009%). This missense change has been observed in individual(s) with male breast cancer (PMID: 20927582). ClinVar contains an entry for this variant (Variation ID: 51658). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is not expected to disrupt BRCA2 protein function with a negative predictive value of 95%. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -
