Uncertain significance, criteria provided, single submitter | clinical testing | Quest Diagnostics Nichols Institute San Juan Capistrano | Jul 02, 2024 | The BRCA2 c.5270A>G (p.Tyr1757Cys) variant has been reported in the published literature in individuals with breast cancer (PMIDs: 35264596 (2022), 28392550 (2018)), suspected Lynch syndrome (PMID: 25980754 (2015)), as well as in reportedly healthy individuals (PMIDs: 35957908 (2022), 33471991 (2021), see also LOVD (http://databases.lovd.nl/shared)). This variant has been described to be located in a region of the BRCA2 gene that is tolerant to missense sequence changes (PMID: 31911673 (2020)). The frequency of this variant in the general population, 0.000012 (3/250310 chromosomes (Genome Aggregation Database, http://gnomad.broadinstitute.org)), is uninformative in the assessment of its pathogenicity. Analysis of this variant using bioinformatics tools for the prediction of the effect of amino acid changes on protein structure and function yielded predictions that this variant is benign. Based on the available information, we are unable to determine the clinical significance of this variant. - |

Hereditary cancer-predisposing syndrome Uncertain:2Benign:1
Uncertain significance, criteria provided, single submitter | clinical testing | Ambry Genetics | Dec 16, 2022 | The p.Y1757C variant (also known as c.5270A>G), located in coding exon 10 of the BRCA2 gene, results from an A to G substitution at nucleotide position 5270. The tyrosine at codon 1757 is replaced by cysteine, an amino acid with highly dissimilar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. - |
Uncertain significance, criteria provided, single submitter | clinical testing | Color Diagnostics, LLC DBA Color Health | Mar 15, 2023 | This missense variant replaces tyrosine with cysteine at codon 1757 of the BRCA2 protein. Computational prediction is inconclusive regarding the impact of this variant on protein structure and function (internally defined REVEL score threshold 0.5 < inconclusive < 0.7, PMID: 27666373). To our knowledge, functional studies have not been reported for this variant. This variant has been reported in two individuals affected with breast cancer (PMID: 28392550, 35264596) and an individual suspected of being affected by Lynch syndrome (PMID: 25980754). This variant has been identified in 3/250310 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance. - |

Uncertain significance, criteria provided, single submitter | clinical testing | Women's Health and Genetics/Laboratory Corporation of America, LabCorp | May 06, 2022 | Variant summary: BRCA2 c.5270A>G (p.Tyr1757Cys) results in a non-conservative amino acid change in the encoded protein sequence. Four of five in-silico tools predict a benign effect of the variant on protein function. The variant allele was found at a frequency of 1.2e-05 in 250310 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.5270A>G has been reported in the literature in at least an individual who underwent clinical genetic testing for Lynch Syndrome and in a breast cancer patient suspected of hereditary breast cancer (Yurgelun_2015, Hwang_2017). These reports do not provide unequivocal conclusions about association of the variant with Hereditary Breast And Ovarian Cancer Syndrome. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. Eight clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014 without evidence for independent evaluation. All laboratories classified the variant as uncertain significance. Based on the evidence outlined above, the variant was classified as uncertain significance. - |

In a large breast cancer case-control study, this variant has been reported in 0/60466 cases and 1/53461 unaffected controls (PMID: 33471991). This variant has been identified in 3/250310 chromosomes in the general population by the Genome Aggregation Database (gnomAD).
