The lysine at codon 1888 is replaced by arginine, an amino acid with highly similar properties. This alteration has been identified in a study of 74 Russian individuals diagnosed with ovarian cancer (Smirnova TY et al. Bull. Exp. Biol. Med., 2007 Jul;144:83-5). Additonally, this alteration was identified in an individual diagnosed with breast cancer (Fanale D et al. Front Oncol, 2021 Jun;11:682445). This variant was also reported in 1/60,466 breast cancer cases and in 1/53,461 controls (Dorling et al. N Engl J Med 2021 02;384:428-439). This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.5663A>G has been reported in the literature in individuals affected with Hereditary Breast And Ovarian Cancer Syndrome (Smirnova_2007, Incorvaia_2020, Fanale_2021). To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. Nine clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014 without evidence for independent evaluation (VUS n=8, likely benign n=1).
