This variant deletes 1 nucleotide in exon 11 of the BRCA2 gene, creating a frameshift and premature translation stop signal. This variant is expected to result in an absent or non-functional protein product. To our knowledge, functional studies have not been performed for this variant. This variant has been reported in two individuals affected with breast cancer (PMID: 16912212, 18703817) and one individual affected with prostate cancer and colon polyps (PMID: 26681312). This variant has been identified in 2/250822 chromosomes in the general population by the Genome Aggregation Database (gnomAD). Loss of BRCA2 function is a known mechanism of disease (clinicalgenome.org). Based on the available evidence, this variant is classified as Pathogenic. -

The BRCA2 c.5828del (p.Ser1943Leufs*20) variant alters the translational reading frame of the BRCA2 mRNA and causes the premature termination of BRCA2 protein synthesis. This variant has been reported in the published literature in individuals and families affected with breast/ovarian cancer (PMID: 18703817 (2008), 22430266 (2012), 25256924 (2014), 30322717 (2018)). It has also been reported in an individual affected with prostate cancer and colon polyps (PMID: 26681312 (2015)), as well as an individual affected with myeloma and acute lymphoblastic leukemia (ALL) (PMID: 33850299 (2021)). The frequency of this variant in the general population, 0.000008 (2/250822 chromosomes (Genome Aggregation Database, http://gnomad.broadinstitute.org)), is consistent with pathogenicity. Based on the available information, this variant is classified as pathogenic. -
Frameshift variant predicted to result in protein truncation or nonsense mediated decay in a gene for which loss-of-function is a known mechanism of disease; Not observed at a significant frequency in large population cohorts (gnomAD); Truncating variants in this gene are considered pathogenic by a well-established clinical consortium and/or database; Also known as 6056delC; This variant is associated with the following publications: (PMID: 29446198, 30720243, 22430266, 18703817, 26681312, 25256924, 30322717, 30787465, 32719484, 29922827, 28888541) -
Hereditary cancer-predisposing syndrome Pathogenic:3
This variant deletes 1 nucleotide in exon 11 of the BRCA2 gene, creating a frameshift and premature translation stop signal. This variant is expected to result in an absent or non-functional protein product. To our knowledge, functional studies have not been performed for this variant. This variant has been reported in two individuals affected with breast cancer (PMID: 16912212, 18703817), an individual affected with ovarian cancer (PMID: 30322717) and an individual affected with prostate cancer (PMID: 26681312). This variant has also been observed in 9 families among the CIMBA participants (PMID: 29446198). This variant has been identified in 2/250822 chromosomes in the general population by the Genome Aggregation Database (gnomAD). Loss of BRCA2 function is a known mechanism of disease (clinicalgenome.org). Based on the available evidence, this variant is classified as Pathogenic. -
The c.5828delC pathogenic mutation, located in coding exon 10 of the BRCA2 gene, results from a deletion of one nucleotide at position 5828, causing a translational frameshift with a predicted alternate stop codon (p.S1943Lfs*20). This alteration has been reported in multiple cohorts of breast and/or ovarian cancer patients (Palma MD et al. Cancer Res. 2008 Sep;68:7006-14; Finkelman BS et al. J. Clin. Oncol. 2012 Apr;30:1321-8; Carter NJ et al. Gynecol Oncol. 2018 12;151:481-488). This alteration was also identified in a patient with prostate cancer and colon polyps (Susswein LR et al. Genet. Med. 2016 08;18:823-32). Of note, this alteration is also designated as 6056delC in published literature. In addition to the clinical data presented in the literature, this alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation. -

Hereditary breast ovarian cancer syndrome Pathogenic:3
Variant summary: BRCA2 c.5828delC (p.Ser1943LeufsX20) results in a premature termination codon, predicted to cause a truncation of the encoded protein or absence of the protein due to nonsense mediated decay, which are commonly known mechanisms for disease. Truncations downstream of this position have been classified as pathogenic by our laboratory. The variant allele was found at a frequency of 7.9e-06 in 252170 control chromosomes (gnomAD and Malone_2006). c.5828delC has been reported in the literature in multiple individuals affected with breast, ovarian, and other cancers, many of whom also have a family history of cancer (e.g. Palma_2008, Finkelman_2012, Malone_2006, Wang_2014, Susswein_2015, Carter_2018, Rebbeck_2018, Singhal_2021, BIC database). These data indicate that the variant is very likely to be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. Nine assessments for this variant have been submitted to ClinVar after 2014 and all classified the variant as pathogenic. Based on the evidence outlined above, the variant was classified as pathogenic. -

This sequence change creates a premature translational stop signal (p.Ser1943Leufs*20) in the BRCA2 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in BRCA2 are known to be pathogenic (PMID: 20104584). This variant is present in population databases (rs80359541, gnomAD 0.0009%). This premature translational stop signal has been observed in individual(s) with breast and/or ovarian cancer (PMID: 18703817, 22430266, 26681312). This variant is also known as 6056delC. ClinVar contains an entry for this variant (Variation ID: 37999). For these reasons, this variant has been classified as Pathogenic. -
