This changes the amino acid from a glutamic acid to a stop codon within coding exon 10. This alteration has been reported in one high-risk breast/ovarian Dutch family (van der Hout AH et al. Hum. Mutat. 2006 Jul;27(7):654-66) and in one Indonesian family with breast cancers diagnosed after age 60 (Purnomosari D et al. Breast Cancer Res Treat (2007) 106:297–304). Of note, this alteration is also referred to as 6775G>T in the published literature. In addition to the clinical data presented in the literature, this alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay.
