This variant deleted 5 basepairs in the intron 11 acceptor site in the BRCA2 gene. Splice site prediction tools predict that this variant may have a significant impact on RNA splicing (PMID: 30661751, 35449021). An RNA study has reported that this variant results in partial in-frame skipping of exon 12, c.6842_6937del (p.Glu2282_Gly2313del) that has been reported to be potentially functional (PMID: 32046981). To our knowledge, this variant has not been reported in individuals affected with BRCA2-related disorders in the literature. This variant has been identified in 2/271982 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance. -
Hereditary breast ovarian cancer syndrome Uncertain:1
This sequence change falls in intron 11 of the BRCA2 gene. It does not directly change the encoded amino acid sequence of the BRCA2 protein. This variant is present in population databases (no rsID available, gnomAD 0.009%). This variant has not been reported in the literature in individuals affected with BRCA2-related conditions. ClinVar contains an entry for this variant (Variation ID: 462416). Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site.
