BRCA2 (HGNC:1101): (BRCA2 DNA repair associated) Inherited mutations in BRCA1 and this gene, BRCA2, confer increased lifetime risk of developing breast or ovarian cancer. Both BRCA1 and BRCA2 are involved in maintenance of genome stability, specifically the homologous recombination pathway for double-strand DNA repair. The largest exon in both genes is exon 11, which harbors the most important and frequent mutations in breast cancer patients. The BRCA2 gene was found on chromosome 13q12.3 in human. The BRCA2 protein contains several copies of a 70 aa motif called the BRC motif, and these motifs mediate binding to the RAD51 recombinase which functions in DNA repair. BRCA2 is considered a tumor suppressor gene, as tumors with BRCA2 mutations generally exhibit loss of heterozygosity (LOH) of the wild-type allele. [provided by RefSeq, May 2020]

It is listed in the dbSNP database as coming from a "clinical source" (ID#: rs4986860) with a MAF score of 0.007 (1000 Genomes). The variant was identified in the ExAc Database at a frequency of 0.003 which includes 9 homozygous individuals and a 0.03 frequency in the African population. The variant was also identified in the UMD (x24), BIC (x79), Exome Server and BOCs databases. In the UMD database, the p.His2440Arg variant was found to co-occur with pathogenic mutations in BRCA1 c.IVS16+6T>C (c.4986+6T>C), as well as in BRCA2 c.2808_2811delACAA (p.Ala938ProfsX21); c.7069_7070delCT (p.Leu2357ValfsX2), increasing the likelihood that the variant is a benign alteration. This residue is not conserved in mammals and computational analyses (SIFT, AlignGVGD) do not suggest a high likelihood of impact to the protein. However, this information is not predictive enough to rule out pathogenicity. In summary, based on the above information, this variant is classified as Benign. -
