The c.7485dupT pathogenic mutation, located in coding exon 14 of the BRCA2 gene, results from a duplication of T at nucleotide position 7485, causing a translational frameshift with a predicted alternate stop codon (p.K2496*). This mutation was identified in 4/1227 African American women with a personal and/or family history of breast, ovarian or gynecologic cancer who underwent germline genetic testing (Barrington DA et al. Gynecol. Oncol., 2018 05;149:337-340). This mutation was also identified in a large, worldwide study of BRCA1/2 mutation positive families (Rebbeck TR et al. Hum. Mutat., 2018 05;39:593-620). This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD). In addition to the clinical data presented in the literature, this alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation. -

The variant was absent in 251448 control chromosomes (gnomAD). The variant, c.7485dupT, has been reported in the literature in individuals affected with Hereditary Breast and Ovarian Cancer (Barrington_2018, Rebbeck_2018). These data indicate that the variant may be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. . Four other ClinVar submissions (evaluation after 2014), including one expert panel (ENIGMA), cite this variant as pathogenic. Based on the evidence outlined above, the variant was classified as pathogenic. -
