The variant was absent in 251316 control chromosomes (gnomAD). c.7543dupA has been reported in the literature in multiple individuals affected with Hereditary Breast And/or Ovarian Cancer and prostate cancer (examples: Kim_2012, Castro_2013, Lalloo_2006, and Edwards_2003). These data indicate that the variant is very likely to be associated with disease. Four submitters including an expert panel (ENIGMA) have submitted clinical-significance assessments for this variant to ClinVar after 2014 and all classified the variant as pathogenic. Breast Cancer Res Treat, 2012 Aug;134:1315-26; Seifert BA et al. Clin Cancer Res, 2016 Aug;22:4087-4094; Dorling et al. N Engl J Med. 2021 02;384:428-439). This mutation has also been detected in multiple prostate cancer patients (Mitra A et al. Br J Cancer, 2008 Jan;98:502-7; Edwards SM et al. Br J Cancer, 2010 Sep;103:918-24; Castro E et al.
