The isoleucine at codon 2828 is replaced by valine, an amino acid with highly similar properties. This alteration was reported in a Portuguese family with breast and ovarian cancer. An additional BRCA2 alteration, p.I2840V, was detected in the same family (Peixoto A et al. Fam. Cancer 2006 ;5(4):379-87). In another study, this alteration was detected in 1/106 high-risk breast and/or ovarian cancer patients from Turkey (Manguolu E et al. Cancer Genet. Cytogenet. 2010 Dec; 203(2):230-7). This alteration was also detected in a cohort of 1663 Brazilian breast cancer patients who underwent hereditary multigene panel testing (Guindalini RSC et al. Sci Rep, 2022 Mar;12:4190). Of note, this alteration is also called 8710A>G in published literature. This amino acid position is poorly conserved in available vertebrate species, and valine is the reference amino acid in several species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. - |
