Variant summary: BRCA2 c.8953+16C>T alters a non-conserved nucleotide located at a position not widely known to affect splicing. Consensus agreement among computation tools predict no significant impact on normal splicing. However, these predictions have yet to be confirmed by functional studies. The variant allele was found at a frequency of 6.1e-05 in 244558 control chromosomes. This frequency is not significantly higher than estimated for a pathogenic variant in BRCA2 causing Hereditary Breast And Ovarian Cancer Syndrome (6.1e-05 vs 0.00075), allowing no conclusion about variant significance. c.8953+16C>T has been reported in the literature in individuals affected with Hereditary Breast And Ovarian Cancer Syndrome (Wagner_1999, Kluska_2015). These report(s) do not provide unequivocal conclusions about association of the variant with Hereditary Breast And Ovarian Cancer Syndrome. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publications have been ascertained in the context of this evaluation (PMID: 25948282, 9971877). ClinVar contains an entry for this variant (Variation ID: 52708).
