This sequence change replaces glycine with arginine at codon 1176 of the ATP7B protein (p.Gly1176Arg). The glycine residue is highly conserved and there is a moderate physicochemical difference between glycine and arginine. This missense change has been observed in individuals with Wilson disease (PMID: 9887381, 11243728, 15845031, 20958917). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt ATP7B protein function.
