Genes affected
DIAPH3 (HGNC:15480): (diaphanous related formin 3) This gene encodes a member of the diaphanous subfamily of the formin family. Members of this family are involved in actin remodeling and regulate cell movement and adhesion. Mutations in this gene are associated with autosomal dominant auditory neuropathy 1. Multiple transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Apr 2012]

Uncertain significance, criteria provided, single submitter | clinical testing | GeneDx | Mar 02, 2018 | The c.3543dupC variant has not been published as a pathogenic variant, nor has it been reported as a benign variant to our knowledge. The variant causes a frameshift starting with codon Glutamic acid 1182, changes this amino acid to an Arginine residue and creates a premature Stop codon at position 3 of the new reading frame, denoted p.Glu1182ArgfsX3. This variant causes the final 12 amino acids to be replaced with 2 incorrect amino acids. However, the variant is observed in 21/18862 (0.11%) alleles from individuals of East Asian background in large population cohorts (Lek et al., 2016). In summary, based on the currently available information, it is unclear whether this variant is a pathogenic variant or a rare benign variant. - |
Uncertain significance, criteria provided, single submitter | clinical testing | Labcorp Genetics (formerly Invitae), Labcorp | Oct 19, 2021 | Experimental studies and prediction algorithms are not available or were not evaluated, and the functional significance of this variant is currently unknown. ClinVar contains an entry for this variant (Variation ID: 504360). This variant has not been reported in the literature in individuals affected with DIAPH3-related conditions. This variant is present in population databases (rs756660986, ExAC 0.08%). This sequence change creates a premature translational stop signal (p.Glu1182Argfs*3) in the DIAPH3 gene. While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 12 amino acid(s) of the DIAPH3 protein. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. - |
