Also referred to as Batten disease, NCL comprises a class of autosomal recessive, neurodegenerative disorders affecting children. The genes responsible likely encode proteins involved in the degradation of post-translationally modified proteins in lysosomes. Lys225Glu is a non-conservative amino acid substitution as a positively charged Lysine residue is replaced by a negatively charged Glutamic acid residue. The variant occurs as a position in the CLN5 protein that is conserved in mammals but is not conserved in more distant species through evolution. Some in silico algorithms predict that Lys225Glu is possibly damaging to the structure/function of the CLN5 protein, while another algorithm predicts that this change is possibly benign. Therefore, based on the currently available information, it is unclear whether Lys225Glu is a disease-causing mutation or a rare benign variant. The variant is found in EPILEPSY panel(s). - |
