The encoded protein can form a homotetramer and has been shown to interact with spastin and with mitogen-activated protein kinase kinase kinase kinase 4. This protein may be involved in axonal maintenance as evidenced by the fact that defects in this gene are a cause of spastic paraplegia type 3. The valine at codon 360 is replaced by methionine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the supporting evidence, this variant is unlikely to be causative of autosomal dominant spastic paraplegia 3A (SPG3A) or hereditary sensory neuropathy type ID (HSN1D); however, its contribution to the development of autosomal recessive spastic paraplegia 3A is uncertain. - |
