The variant allele was found at a frequency of 0.00054 in 250802 control chromosomes, predominantly at a frequency of 0.0071 within the East Asian subpopulation in the gnomAD database. To our knowledge, no occurrence of c.443G>C in individuals affected with Hereditary Spastic Paraplegia and no experimental evidence demonstrating its impact on protein function have been reported. Three submitters have cited clinical-significance assessments for this variant to ClinVar after 2014. Two submitters classified the variant as uncertain significance and one classified it as benign.
