Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 1.6e-05 in 251494 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.1270G>T has been reported in the literature as a non-informative genotype (second allele and/or zygosity not specified) in cohorts of individuals affected with Congenital Hypothyroidism (example, Long_2018, Wang_2020, Xue_2021, Li_2023). These report(s) do not provide unequivocal conclusions about association of the variant with Hypothyroidism Due To TSH Receptor Mutations. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publications have been ascertained in the context of this evaluation (PMID: 36125728, 30022773, 32425884, 34377013).
