Other variant(s) that disrupt this residue have been determined to be pathogenic (PMID: 21070211). This suggests that this residue is clinically significant, and that variants that disrupt this residue are likely to be disease-causing. This variant has not been reported in the literature in individuals affected with GALC-related conditions. The frequency data for this variant in the population databases is considered unreliable, as metrics indicate insufficient coverage at this position in the ExAC database. This sequence change replaces glycine with cysteine at codon 57 of the GALC protein (p.Gly57Cys). The glycine residue is highly conserved and there is a large physicochemical difference between glycine and cysteine. -
