The encoded protein functions as a ribonuclease and is required by the RNA interference and small temporal RNA (stRNA) pathways to produce the active small RNA component that represses gene expression. This protein also acts as a strong antiviral agent with activity against RNA viruses, including the Zika and SARS-CoV-2 viruses. Alternative splicing results in multiple transcript variants. The normal sequence, with the bases that are deleted in brackets, is GACT[delTT]GTGC. The deletion causes a frameshift which changes a Phenylalanine to a Cysteine at codon 1772, and creates a premature stop codon at position 6 of the new reading frame. This variant is predicted to cause loss of normal protein function through either protein truncation or nonsense-mediated mRNA decay. DICER1 c.5315_5316delTT has been reported in at least one individual with pleuropulmonary blastoma (Brenneman 2015). We consider this variant to be pathogenic. - |
