In summary, this is a novel missense change with uncertain impact on protein function. It has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function do not agree on the potential impact of this missense change (SIFT: "Tolerated"; PolyPhen-2: "Possibly Damaging"; Align-GVGD: "Class C0"). This variant is not present in population databases (ExAC no frequency) and has not been reported in the literature. This sequence change replaces serine with isoleucine at codon 1491 of the DICER1 protein (p.Ser1491Ile). The serine residue is weakly conserved and there is a large physicochemical difference between serine and isoleucine. -
Hereditary cancer-predisposing syndrome Uncertain:1
The p.S1491I variant (also known as c.4472G>T), located in coding exon 22 of the DICER1 gene, results from a G to T substitution at nucleotide position 4472. The serine at codon 1491 is replaced by isoleucine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
