Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt DICER1 protein function with a negative predictive value of 80%. Hereditary cancer-predisposing syndrome Uncertain:1
The p.L370P variant (also known as c.1109T>C), located in coding exon 7 of the DICER1 gene, results from a T to C substitution at nucleotide position 1109. The leucine at codon 370 is replaced by proline, an amino acid with similar properties. This alteration has been identified in an individual with a personal history of ovarian cancer and a maternal family history of colon cancer (Chan GHJ et al. Oncotarget, 2018 Jul;9:30649-30660). This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
