Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 251406 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.1157G>A has been reported in the literature in a heterozygous individual affected with features of Alpers' syndrome (Baruffini_2011). This report does not provide unequivocal conclusions about association of the variant with Mitochondrial DNA Depletion Syndrome - POLG Related. This publication reports experimental evidence evaluating an impact on protein function in yeast, finding that replacing an Arg with a His residue in a humanized yeast strain results in a higher frequency of the petite phenotype. However, this does not allow convincing conclusions about the variant effect. The following publication has been ascertained in the context of this evaluation (PMID: 20883824).
