The p.E1232Q variant (also known as c.3694G>C), located in coding exon 27 of the MYH11 gene, results from a G to C substitution at nucleotide position 3694. The glutamic acid at codon 1232 is replaced by glutamine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear. -

This sequence change replaces glutamic acid, which is acidic and polar, with glutamine, which is neutral and polar, at codon 1239 of the MYH11 protein (p.Glu1239Gln). This variant is present in population databases (rs754761681, gnomAD 0.002%). This variant has not been reported in the literature in individuals affected with MYH11-related conditions. ClinVar contains an entry for this variant (Variation ID: 465728).
