To our knowledge, this variant has not been reported in the literature or in a large population database (http://gnomad.broadinstitute.org), indicating this variant is rare. Of note, a different substitution at the same codon defined as c.2540T>C (p.Leu847Pro) has been reported in individuals with tuberous sclerosis (Supplementary Table 1 of Ding et al. 2020. PubMed ID: 32211034; Li et al. 2011. PubMed ID: 21811971). At this time, the clinical significance of the p.Leu847Val variant is uncertain due to the absence of conclusive functional and genetic evidence. - |
