The p.P1314S variant (also known as c.3940C>T), located in coding exon 32 of the TSC2 gene, results from a C to T substitution at nucleotide position 3940. The proline at codon 1314 is replaced by serine, an amino acid with similar properties. This variant was identified in a cohort of 9769 individuals with epilepsy undergoing genetic testing (Truty R et al. Epilepsia Open, 2019 Sep;4:397-408). This amino acid position is not well conserved in available vertebrate species, and serine is the reference amino acid in other vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear. -
