ClinVar contains an entry for this variant (Variation ID: 565643). This missense change has been observed in individual(s) with epilepsy (PMID: 31875159). This variant is not present in population databases (gnomAD no frequency). This sequence change replaces glutamic acid, which is acidic and polar, with glutamine, which is neutral and polar, at codon 1476 of the TSC2 protein (p.Glu1476Gln). -
Hereditary cancer-predisposing syndrome Uncertain:1
The p.E1476Q variant (also known as c.4426G>C), located in coding exon 33 of the TSC2 gene, results from a G to C substitution at nucleotide position 4426. The glutamic acid at codon 1476 is replaced by glutamine, an amino acid with highly similar properties. This alteration was identified in 1 of 243 epilepsy patients undergoing whole exome sequencing (Kang KW et al. PeerJ, 2019 Dec;7:e8278). This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
