This sequence change replaces glycine, which is neutral and non-polar, with serine, which is neutral and polar, at codon 853 of the PALB2 protein (p.Gly853Ser). This variant is present in population databases (rs587782579, gnomAD 0.02%). This variant has not been reported in the literature in individuals affected with PALB2-related conditions. ClinVar contains an entry for this variant (Variation ID: 142600). The p.G853S variant (also known as c.2557G>A), located in coding exon 6 of the PALB2 gene, results from a G to A substitution at nucleotide position 2557. The glycine at codon 853 is replaced by serine, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
This missense variant replaces glycine with serine at codon 853 of the PALB2 protein. Computational prediction suggests that this variant may not impact protein structure and function (internally defined REVEL score threshold <= 0.5, PMID: 27666373). To our knowledge, functional studies have not been reported for this variant. This variant has been reported in one individual affected with prostate cancer (PMID: 28259476). This variant has been identified in 7/282682 chromosomes in the general population by the Genome Aggregation Database (gnomAD).
