This particular variant has been reported in the literature in an individual affected with breast cancer (PMID: 27616075). This sequence change deletes 1 nucleotide from exon 4 of the PALB2 mRNA (c.1046delA), causing a frameshift at codon 349. This creates a premature translational stop signal (p.Asn349Ilefs*7) and is expected to result in an absent or disrupted protein product. -

The c.1046delA pathogenic mutation, located in coding exon 4 of the PALB2 gene, results from a deletion of one nucleotide at nucleotide position 1046, causing a translational frameshift with a predicted alternate stop codon (p.N349Ifs*7). This alteration has been reported in a individual with a personal and family history of breast cancer (Kraus C et al. Int. J. Cancer, 2017 Jan;140:95-102). In addition to the clinical data presented in the literature, this alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation. -
