This sequence change replaces glycine, which is neutral and non-polar, with arginine, which is basic and polar, at codon 196 of the MEFV protein (p.Gly196Arg). This variant is present in population databases (rs104895179, gnomAD 0.005%). This missense change has been observed in individual(s) with clinical features of MEFV-related conditions (PMID: 26215181). ClinVar contains an entry for this variant (Variation ID: 651794). Algorithms developed to predict the effect of missense changes on protein structure and function output the following: SIFT: "Tolerated"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0".
