It functions as a DNA-binding transcription factor by using distinct zinc fingers in different signaling pathways. Thus, it is thought that this gene may have multiple roles in signal transduction during development. Mutations in this gene are associated with nephronophthisis-14 and Joubert syndrome-19. This variant is present in population databases (rs200585917, gnomAD 0.03%). This missense change has been observed in individual(s) with nephronophthisis (PMID: 22863007). ClinVar contains an entry for this variant (Variation ID: 37287). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) has been performed at Invitae for this missense variant, however the output from this modeling did not meet the statistical confidence thresholds required to predict the impact of this variant on ZNF423 protein function. Experimental studies have shown that this missense change does not substantially affect ZNF423 function (PMID: 22863007, 32925911).
