Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 4e-06 in 249394 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.1871C>T in individuals affected with Fanconi Anemia has been reported. A publication reported experimental evidence evaluating interactions of the variant protein with HSP70 and HSP90, but this assay does not allow convincing conclusions about the variant effect on protein function (Karras_2017). Two other clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014, and both of them classified the variant as uncertain significance.
