This variant is also absent from general population databases (Exome Variant Server, Genome Aggregation Database), indicating it is not a common polymorphism. This variant induces an early termination codon and is predicted to result in a truncated protein or mRNA subject to nonsense-mediated decay. Truncating variants downstream of codon 851 have been reported in numerous individuals and families with neurofibromatosis and are considered pathogenic (Sabbagh 2013, van Minkelen 2014), supporting the notion that the p.Cys851Ter variant is deleterious.
