The NF1 c.2585C>G (p.Thr862Ser) variant has been reported in the published literature in individuals affected with Neurofibromatosis Type 1 (NF1) (PMID: 23656349 (2014), 29290338 (2017), 30308447 (2018)). The frequency of this variant in the general population, 0.000062 (8/128702 chromosomes (Genome Aggregation Database, http://gnomad.broadinstitute.org)), is uninformative in the assessment of its pathogenicity. Analysis of this variant using bioinformatics tools for the prediction of the effect of amino acid changes on protein structure and function yielded conflicting predictions that this variant is deleterious or benign. Based on the available information, we are unable to determine the clinical significance of this variant. -

Thep.T862Svariant (also known as c.2585C>G), located in coding exon 21 of theNF1gene, results from a C to G substitution at nucleotide position 2585. The threonine at codon 862 is replaced by serine, an amino acid with similar properties. This variant was previously reported in the SNPDatabase as rs200302954, but was absent from population-based cohorts in the NHLBI Exome Sequencing Project (ESP) and 1000 Genomes Project databases.To date, this alteration has been detected with an allele frequency of approximately 0.01% (greater than 110000 alleles tested) in our clinical cohort.This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance ofp.T862Sremains unclear. -
