The c.2728_2729delGG pathogenic mutation, located in coding exon 21 of the NF1 gene, results from a deletion of two nucleotides at nucleotide positions 2728 to 2729, causing a translational frameshift with a predicted alternate stop codon (p.G910Tfs*8). This alteration was identified in an individual diagnosed with neurofibromatosis type 1 (NF1) (Wang X et al. Genes Chromosomes Cancer, 2018 Jan;57:19-27). This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD). In addition to the clinical data presented in the literature, this alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation. -
