This variant has been observed to be de novo in several individuals affected with neurofibromatosis type 1 (PMID: 26740943, 21520333,  Invitae). ClinVar contains an entry for this variant (Variation ID: 527596). This variant is not present in population databases (ExAC no frequency). This sequence change replaces methionine with arginine at codon 1215 of the NF1 protein (p.Met1215Arg). The methionine residue is moderately conserved and there is a moderate physicochemical difference between methionine and arginine. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Possibly Damaging"; Align-GVGD: "Class C0"). For these reasons, this variant has been classified as Pathogenic. This variant disrupts the p.Met1215 amino acid residue in NF1. Other variant(s) that disrupt this residue have been observed in individuals with NF1-related conditions (PMID:10712197, 16835897, 18546366 ), suggesting that it is a clinically significant residue. As a result, variants that disrupt this residue are likely to be causative of disease.
