The lysine residue is moderately conserved and there is a small physicochemical difference between lysine and glutamic acid. This variant is not present in population databases (ExAC no frequency). This variant has been reported in the literature in an individual suspected with neurofibromatosis. However, this individual did not meet clinical criteria for the disease (PMID: 15060124). Family studies indicate this missense variant likely was not inherited from either parent and occurred de novo in proband with multiple cafe au lait spots, neurofibromas, and lisch nodules. In summary, this variant has not been observed in the population databases, but observed in individuals with suspected neurofibromatosis. It was confirmed to have arisen de novo through family testing in an affected individual. For these reasons, this variant has been classified as Pathogenic. -
