The variant of interest was observed in the large, broad control population, ExAC, with an allele frequency of 699/109636 (21 homozygotes, 1/156, frequency: 0.0063756), predominantly observed in the African cohort, 660/9614 (21 homozygotes, 1/14, frequency: 0.06865), which significantly exceeds the estimated maximal expected allele frequency for a pathogenic NF1 variant of 1/4798 (0.0002084). Therefore, suggesting the variant is a common polymorphism found in population(s) of African origin. The variant of interest, to our knowledge, has not been reported in affected individuals via publications and/or reputable databases/clinical laboratories.
