The RAD51D c.911G>A (p.Gly304Asp) missense change has a maximum subpopulation frequency of 0.0062% in gnomAD v2.1.1 (https://gnomad.broadinstitute.org/). The in silico tool REVEL predicts a benign effect on protein function, but to our knowledge this prediction has not been confirmed by functional studies. This variant was identified 1 of 911 individuals with personal and family history of breast and/or ovarian cancer and in 0 of 1060 population controls (PMID: 21822267). It is absent in a database of women older than 70 years of age who have never had cancer (FLOSSIES database, https://whi.color.com/). To our knowledge, this variant has not been reported in individuals with Fanconi anemia. In summary, the evidence currently available is insufficient to determine the clinical significance of this variant. It has therefore been classified as of uncertain significance. -
This sequence change replaces glycine, which is neutral and non-polar, with aspartic acid, which is acidic and polar, at codon 304 of the RAD51D protein (p.Gly304Asp). This variant is present in population databases (rs200615280, gnomAD 0.006%). This variant has not been reported in the literature in individuals affected with RAD51D-related conditions. ClinVar contains an entry for this variant (Variation ID: 141707). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is not expected to disrupt RAD51D protein function with a negative predictive value of 80%. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -

In silico analysis supports that this missense variant has a deleterious effect on protein structure/function; Observed in individuals with a personal or family history of breast or ovarian cancer, as well as unaffected control groups, and a father and son both with Hodgkin lymphoma from a single family (PMID: 21822267, 35977101, 33471991); This variant is associated with the following publications: (PMID: 35977101, 21822267, 33471991, 21111057, 14704354, 19327148) -

Hereditary cancer-predisposing syndrome Uncertain:2
The p.G304D variant (also known as c.911G>A), located in coding exon 10 of the RAD51D gene, results from a G to A substitution at nucleotide position 911. The glycine at codon 304 is replaced by aspartic acid, an amino acid with similar properties. In one study, this alteration was detected in 1/911 breast-ovarian cancer families but not in 1060 population controls (Loveday C et al. Nat. Genet. 2011;43:879-82). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
This missense variant replaces glycine with aspartic acid at codon 304 of the RAD51D protein. Computational prediction suggests that this variant may not impact protein structure and function. To our knowledge, functional studies have not been reported for this variant. This variant has been reported in an individual affected with breast cancer (PMID: 33471991), an individual that met criteria for hereditary breast and ovarian cancer syndrome (PMID: 21822267), and a family affected with Hodgkin lymphoma (PMID: 35977101). This variant has also been identified in 8/282882 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance. -
not specified Uncertain:1
Variant summary: RAD51D c.911G>A (p.Gly304Asp) results in a non-conservative amino acid change in the encoded protein sequence. Three of five in-silico tools predict a benign effect of the variant on protein function. The variant allele was found at a frequency of 2e-05 in 253602 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.911G>A has been reported in the literature in individuals affected with Breast/Ovarian Cancer without strong evidence for causality (e.g. Loveday_2011, Dorling_2021). These reports do not provide unequivocal conclusions about association of the variant with Hereditary Breast And Ovarian Cancer Syndrome. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. Five ClinVar submitters have assessed the variant since 2014: all five classified the variant as uncertain significance. Based on the evidence outlined above, the variant was classified as uncertain significance. -
