Breast-ovarian cancer, familial, susceptibility to, 4 Pathogenic:4Other:1
This variant is considered pathogenic. This variant creates a frameshift predicted to result in premature protein truncation. -
- -
This sequence change creates a premature translational stop signal (p.Val28Trpfs*12) in the RAD51D gene. RNA analysis indicates that this premature translational stop signal induces altered splicing and is likely to result in the loss of the initiator methionine. This variant is not present in population databases (gnomAD no frequency). This premature translational stop signal has been observed in individual(s) with ovarian cancer (PMID: 26720728). ClinVar contains an entry for this variant (Variation ID: 419558). Studies have shown that this premature translational stop signal results in skipping of exon 1, and is expected to result in the loss of the initiator methionine (internal data). For these reasons, this variant has been classified as Pathogenic. -
Variant interpreted as Pathogenic and reported on 07-08-2018 by Lab Invitae. GenomeConnect-Invitae Patient Insights Network assertions are reported exactly as they appear on the patient-provided report from the testing laboratory. Registry team members make no attempt to reinterpret the clinical significance of the variant. Phenotypic details are available under supporting information. -

not provided Pathogenic:2
This variant alters the translational reading frame of the RAD51D mRNA and causes the premature termination of RAD51D protein synthesis. In the published literature, this variant has been reported in individuals affected with ovarian cancer (PMID: 26720728 (2016)) or prostate cancer (PMID: 31948886 (2020)). This variant has not been reported in large, multi-ethnic general populations (Genome Aggregation Database, http://gnomad.broadinstitute.org). Based on the available information, this variant is classified as pathogenic. -
Frameshift variant predicted to result in protein truncation or nonsense mediated decay in a gene for which loss of function is a known mechanism of disease; Not observed at significant frequency in large population cohorts (gnomAD); This variant is associated with the following publications: (PMID: 26720728, 31589614, 31948886, 31575519) -
Hereditary cancer-predisposing syndrome Pathogenic:2
This variant deletes 1 nucleotide in exon 1 of the RAD51D gene, creating a frameshift and premature translation stop signal. This variant is expected to result in an absent or non-functional protein product. This variant has been reported in an individual affected with ovarian cancer (PMID: 26720728) and in an individual affected with breast cancer (PMID: 31575519). This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). Loss of RAD51D function is a known mechanism of disease (clinicalgenome.org). Based on the available evidence, this variant is classified as Pathogenic. -
The c.81delA pathogenic mutation, located in coding exon 1 of the RAD51D gene, results from a deletion of one nucleotide at nucleotide position 81, causing a translational frameshift with a predicted alternate stop codon (p.V28Wfs*12). This alteration was identified in an ovarian cancer patient (Norquist BM et al. JAMA Oncol. 2016 Apr;2(4):482-90). This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD). In addition to the clinical data presented in the literature, this alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation. -
