The Ala101Thr variant in TCAP has been previously observed in our laboratory in one individual with a clinical diagnosis of ARVC. Data from large population st udies is insufficient to assess the frequency of this variant. Computational an alyses (biochemical amino acid properties, conservation, AlignGVGD, PolyPhen2, a nd SIFT) do not provide strong support for or against an impact to the protein. In summary, additional information is needed to fully assess the clinical signi ficance of the Ala101Thr variant. -
