This missense variant replaces methionine with threonine at codon 1137 of the BRCA1 protein. Computational prediction suggests that this variant may not impact protein structure and function. To our knowledge, functional studies have not been reported for this variant. This variant has been reported in an individual affected with breast cancer (PMID: 19491284) and in an individual with a family history of breast and/or ovarian cancers (PMID: 22476429). This variant also has been reported in one ore more individuals homozygous for this variant without expected clinical features if this variant were disease-causing (ClinVar SCV004186056.1). This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance. -
The p.M1137T variant (also known as c.3410T>C), located in coding exon 9 of the BRCA1 gene, results from a T to C substitution at nucleotide position 3410. The methionine at codon 1137 is replaced by threonine, an amino acid with similar properties. This alteration has been identified in high-risk breast/ovarian cohorts (Haffty BG et al. J. Med. Genet. 2006 Feb;43(2):133-7; Lu W et al. Fam Cancer, 2012 Sep;11:381-5). This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -

Variant summary: BRCA1 c.3410T>C (p.Met1137Thr) results in a non-conservative amino acid change in the encoded protein sequence. Four of five in-silico tools predict a benign effect of the variant on protein function. The variant was absent in 250962 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.3410T>C has been reported in the literature as a VUS in individuals affected with Breast and/or Ovarian Cancer (e.g. Haffty_2006, Lu_2012). These reports do not provide unequivocal conclusions about association of the variant with Hereditary Breast and Ovarian Cancer. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publications have been ascertained in the context of this evaluation (PMID: 16518693, 12531920, 19491284, 15983021, 22476429, 15385441, 30212499, 23704879). Six submitters have cited clinical-significance assessments for this variant to ClinVar after 2014. Five submitters classified the variant as uncertain significance, and one submitter classified the variant as likely benign. Based on the evidence outlined above, the variant was classified as uncertain significance. -
