The BRCA1 c.2281G>C (p.Glu761Gln) variant has been reported in the published literature in multiple individuals with breast and/or ovarian cancer (PMID: 8875986 (1996), 30254663 (2018)). This variant has been seen where an alternate explanation for disease was also identified, suggesting this variant may not cause disease (PMID: 32438681 (2020)). In the published literature, this variant has been reported to be located in a region of the BRCA1 gene that is tolerant to missense sequence changes (PMID: 31911673 (2020)). The frequency of this variant in the general population, 0.000008 (2/251170 chromosomes (Genome Aggregation Database, http://gnomad.broadinstitute.org)), is uninformative in the assessment of its pathogenicity. Analysis of this variant using bioinformatics tools for the prediction of the effect of amino acid changes on protein structure and function yielded conflicting predictions that this variant is benign or damaging. Based on the available information, we are unable to determine the clinical significance of this variant. -

The p.E761Q variant (also known as c.2281G>C), located in coding exon 9 of the BRCA1 gene, results from a G to C substitution at nucleotide position 2281. The glutamic acid at codon 761 is replaced by glutamine, an amino acid with highly similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
