This sequence change replaces serine, which is neutral and polar, with tryptophan, which is neutral and slightly polar, at codon 163 of the CHRNE protein (p.Ser163Trp). This variant is present in population databases (rs121909516, gnomAD 0.04%). This variant has not been reported in the literature in individuals affected with CHRNE-related conditions. ClinVar contains an entry for this variant (Variation ID: 1038740). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is expected to disrupt CHRNE protein function with a positive predictive value of 95%. This variant disrupts the p.Ser163 amino acid residue in CHRNE. Other variant(s) that disrupt this residue have been determined to be pathogenic (PMID: 8755487).
